過敏原原料的生產製造

Citeq以蟎蟲作為原料來製備數種萃取物,例如有些產品是從整批的培養物或蟎體中萃取,以明確獲得冷凍乾燥萃取物。只要確保其一致性,這些原料跟冷凍乾燥萃取物均可用於診斷、研究和免疫治療目的。Citeq根據明確定義的參數來生產產品,這些參數都有受到監控和記錄,得以維持產品的品質始終如一以滿足客戶的需求標準。

 

生產控制

要從原料中獲得可被接受並且盡可能達到最高標準純度的冷凍乾燥萃取物,其生產過程需要包括幾個重點。首先,最重要的一點,是在萃取過敏原蛋白質時,對應起始材料選用合適的緩衝液,並且在萃取過程中將環境控制在適當的溫度。接著,透過離心跟過濾步驟,將原料溶液中的過敏原蛋白跟不可溶性物質分離開來。

 

品質一致

為了確保萃取物的品質一致,只會從明確的培養基培養出來的培養物上取得產品原料。Citeq 已經為產品來源的每個蟎物種建立了最佳的培養環境與條件,以利作為原料來源的蟎蟲作最佳化的繁殖生長,並在此操控下最有效率地提高原料中的主要過敏原含量。

在理想狀況下,Citeq 能為過敏原產品提供其生物活性的量化數據。可惜的是,現有的科學技術尚無法達到這個目標。因此為了盡量確保生產過程的品質,Citeq 制定了生產規格的參數,並設置監控和記錄來確保品質的一致。

House Dust Mites Allergens

House dust mites are arachnoids and are present in every household and are living within dust, textile, carpets, beds and upholster furniture. HDMs belong to the Pyroglyphidae family. The most important House dust mites are Dermatophagoides pteronyssinus and Dermatophagoides farinae (in drier areas). HDMs contain allergens which is a (non) protein or substance which can induce an allergic response. Humans mostly get exposed to house dust mites and other indoor allergens. Humans who are atopic can react on this exposure by developing an allergy. Allergic responses can result in eczema, conjunctivitis, asthma symptoms and rhinitis.

Our main focus is the production of house dust mites (HDMs) and their allergens. Citeq was born based on a passion for mites and the research for how mites influence our daily lives. Our founders and leading scientists have pharmaceutical, biological and biochemical backgrounds. With over 25 years of experience and an extensive knowledge about mites Citeq is more than just a supplier of allergen source material. Our specialty starts with our cultivation medium which has an absolute low in foreign protein. Our cultivation technique leads to high concentrations of the major allergens, resulting in strong raw materials, extracts in several grades of purity, and very pure major allergens. All our house dust mite products contain the native proteins, with no denaturing treatments during production and purification

Storage Mites

Traditionally Citeq has cultivated 8 species of acarid storage mites (stored product mites) in the families Acaridae, Glycphagidae and Pyroglyphidae. We can deliver the following storage mites: Blomia tropicalis and Acarus Siro.

For research purposes we can also deliver Glycyphagus domesticus, Lepidoglyphus destructor, Tyrophagus putrescentiae, Lepidoglyphus fustifer, Austroglycyphagus geniculatus and Chortoglyphus arcuatus.

Stored product mites are arachnoids and are present in stored food like dried fruits, flour and cereal products. Besides that they can also be found outdoors within hay and straw. The most common species are Blomia tropicalis, Acarus Siro, Glycyphagus domesticus and Lepidoglyphus destructor. Storage mites are a possible cause of allergic disorders and mostly present among agriculture population. Storage mites can also adapt their feeding to house dust. Therefore, they can also cause allergy in more urban areas where the mites life indoor.

Molds (Fungi)

Molds belong to the kingdom Fungi. Molds reproduce by releasing “seeds”, called spores, in the air. These spores contain allergens. Inhaling these spores lead, in atopic human, to an allergic reaction. Spores spread mainly from spring to early fall. Due to the fact that fungi also lives indoor, allergic reactions will occur throughout the entire year. It is estimated that 3-10% of the population, worldwide, is sensitized to mold. From all of the mold species only a few can cause allergic reactions. One of these species is the Alternaria alternata (also know as Alternaria tenuis) which is abundantly present indoor and outdoor. A. alternata has allergens which contains both mycelial fragments and airborne spores which can cause allergic symptoms. Humans who suffer from mold allergy have an IgE reactivity roundabout 70% to Alternaria. A mold allergy can cause a severe respiratory reaction to persons with allergic bronchitis/ asthma.

Cockroach

Cockroaches contain one of the major indoor allergens, inducing respiratory allergies. Cockroaches can be found in large numbers worldwide, this makes cockroaches a cosmopolitan. Worldwide there life about 3500 different cockroach species. The five cockroaches which are known to induce respiratory allergies include Blattella germanica, Blatta orientalis, Periplaneta americana, Supella longipalpis and Periplaneta fuliginosa. Sensitisation to these species in asthma patients varies between 30-60% worldwide. Blattella germanica is a common specie all over the world, therefore, B. germanica is often found in sensitisation profiles of patients with respiratory allergy.

Bee and wasp venoms

Hymenoptera consist of different families, including the families Apidae and Vespidae. Three species which are very important in the context of venom allergies are Vespula vulgaris (the common wasp), Vespula germanica (german wasp) and Apis mellifera (honey bee). Stings of Hymenoptera occur very frequently. 0.3-7.5% of the population, worldwide, suffer from venom allergy. When humans, with a venom allergy, get stung they will suffer from severe local reactions, systemic symptoms. Atopic humans will suffer from an anaphylactic schock after a sting. In central Europe, 30-40% of the human with a venom allergy have an allergy for both, bee and wasp, venoms.

Biotinylated products

During biotinylation biotin is attached to macromolecules or proteins. Biotinylated proteins can, due to the high specifity of biotin-avidin interaction, be used to detect and/ or purify proteins. Citeq has developed house dust mite related biotinylated proteins. These biotinylated proteins include D. pteronyssinus (HDM) extract and the natural purified Der p 1 protein and Der p 2 protein. Biotinylated proteins can be applicated in many protein assays. These protein assays include for example western blot analysis, fluorescence-activated cell sorting (FACS)/ flow cytometry and enzyme-linked immunosorbant assay (ELISA).

anti-mouse antibodies

Together with our biotinylated HDM allergen products, antibodies can be used to quantify HDM allergen specific IgE/ IgG1/ IgG2a antibodies in mouse serum using an ELISA method. The antibody can be coated on an ELISA plate and act as a capture antibody to immobilize serum IgE/ IgG1/ IgG2a antibodies. In the next step biotinylated HDM allergens are added to facilitate the detection of HDM allergen specific antibodies of the IgE/ IgG1/ IgG2a isotype. Using a reference serum or by comparing multiple sera the HDM allergen specific antibodies can be (relatively) quantified. A perfect way for researchers, who are working on allergic asthma research with mouse models, to determine whether immunizations of mice with house dust mite (HDM) have been successful by looking at the amount of specific antibodies against HDM proteins in the serum of mice.
