CRM197 and TTHc

EcoCRM™

EcoCRM™, economical CRM from E. coli, will significantly reduce the cost of conjugate vaccines.
CRM197, a genetically detoxified diphtheria toxin, is widely used as a carrier protein in conjugate vaccines. Effective conjugate vaccines against Streptococcus pneumonaieHaemophilus influenzae b and Neisseria meningitidis have been made using this carrier protein. Fina Biosolutions has developed a highly efficientEscherichia coli expression system for CRM197, along with a simple purification scheme.  We express soluble, properly folded CRM197 in the cytoplasm of E.coli and achieve expression yields of grams per liter.

Recombinant Tetanus Toxin Heavy Chain Fragment C (rTTHc)

Tetanus toxoid (TT) is frequently used as the carrier protein for conjugate vaccines, in addition to being a component of the DPT vaccine. TT contains very strong T cell epitopes. However, as a toxoided protein, many of the surface lysines are blocked by the toxoiding process. TT is not a uniform product. Each manufacturer has its own specific toxoiding and purification process. Tetanus tends to aggregate with age. Furthermore, TT is not generally affordable in the quantities needed for research and early clinical work.

Tetanus toxoid heavy chain fragment C (TTHc) is the C terminal fragment of the toxin. It is devoid of the toxin’s enzymatic activity but contains the binding site necessary for uptake of the toxin by neurons. Significantly, the majority of the antibody response to tetanus toxoid is against epitopes contained within TTHc. Fragment C contains the powerful T cell epitopes of tetanus toxin without the toxicity.

Fina Biosolutions now offers recombinant TTHc (rTTHc), combining the advantages of the toxoid with the reproducibility of a recombinant protein. FinaBio expresses rTTHc as a properly folded, soluble protein in E. coli cytoplasm.

Amino-Dextran

Amino-Dextran (AmDex, AECM-dextran) consists of dextran functionalized with primary amines. Our AmDex is prepared from dextran using a protocol yielding a stable, amino functionalized dextran that is superior to the more commonly used reductive amination methods. The molecular weight profile of Fina Biosolutions’ AmDex is essentially unchanged from the starting dextran, as determined by analytical size exclusion chromatography. The product is desalted and lyophilized. We do extensive processing and testing to ensure that free amino reagent is removed, resulting in a consistent product with minimal lot-to-lot variation.

We have found that many commercial lots of amino-dextran contain variable amounts of dextran and that the amine content is often incorrect. Furthermore, the actual polymer molecular weight distributions of these preparations can vary extensively from the indicated molecular weight. These issues can mean that optimization data is based on incorrect information. Lot-to-lot variations means that protocols may need to be revised when a new lot of AmDex is used.

We take care to ensure that we provide a consistent product. Each batch of amino-dextran polymer is evaluated for molecular weight distribution and for the amine content, expressed as amines/polymer as well as assayed to determine the weight percent dextran. A certificate of analysis is provided for each lot.
Amino-dextrans are available in molecular weights from 3.5 kDa to 2000 kDa in a variety of amine ratios. Custom polymers can be synthesized to match your needs.

Amino-Ficoll

As a T-cell independent carrier (TI carrier), coupling Ficoll with haptens has been shown to produce robust antigen-specific primary immune memory. Our method for amino functionalization yields stable primary amine groups. We carefully control the amine:polymer ratio and process to remove excess amine reagent. Custom Ficoll preparations are available upon request. GE Healthcare’s trademark Ficoll is a highly branched, 400kDa, hydrophilic polysaccharide widely used for blood separation.

Antibody-Dextran Conjugates

Crosslinking membrane IgD on B cells with unconjugated anti-IgD antibody induces B cell proliferation at µg/ml concentrations. By contrast, anti-IgD dextran induces vigorous B cell proliferation at ng/ml. Furthermore, in contrast to unconjugated anti-IgD, anti-IgD-dex-activated B cells undergo substantial Ig class switching and differentiation into Ig-secreting cells in the presence of various second signals, including cytokines and Toll-like receptor ligands. Thus, Anti-IgD-dex is an efficient and potent polyclonal B cell activator for studying a wide range of B cell functions.
Fina Biosolutions provides anti-IgD and anti-IgM dextran conjugates, targeting mouse and human.
Streptavidin-dextran can be used to prepare multivalent polymer conjugates using a biotinylated species (i.e., antibody, TCR, antigen, etc.)

Mono-functionalized Dextrans

Monoamino dextrans and monocarboxyl dextrans are prepared by proprietary methods yielding a stable, monofunctionalized dextran, with a single functional group on the end of the polymer. The molecular weight profile is essentially unchanged, as determined by analytical size exclusion chromatography. Extensive processing and testing is performed to ensure that nonpolymer reagent is removed. This approach results in a consistent product with minimal lot-to-lot variation.
Monothiol dextran is synthesized with a single protected thiol (Dithiolpyridyl group).
These monofunctionalized dextrans can be used much like PEG—it can be used to increase protein half-life, reduce immunogenicity, and passivate surfaces. They can be labeled with a single fluorescent group to make a well-defined tracer.

Specialty immunoreagents

Fina Biosolutions’ Specialty Immunoreagents are a collection of reagents mainly of interest to immunologists. These reagents arose out of both our own research and requests from other scientists. Please let us know if there is a reagent you would like us to make.

TNP-Dextran and TNP-Ficoll are classic T-cell independent model antigens. Due to the wide available of TNP immunoreagents, they serve as useful models of natural T-cell independent antigens like polysaccharides. TNP-OVA and TNP-BSA serve as T-cell dependent model antigens with the trinitrophenyl epitope. The set of TNP reagents also allows for the selection of a suitable ELISA antigen.

Our phosphorylcholine (PC) conjugates are based on the method of Chesboro and Metzger utilizing p-diazonium phenylphosphorylcholine.
Biotin labeled heparin is used in the identification of heparin binding sites. Our heparin conjugation utilizes the terminal end of heparin polymer resulting in minimum modification of the heparin.
